XX XX In developing countries, cancer of the cervix is responsible for XX 24% of all cancers in women. In these areas, it is the most XX frequent female malignancy. In developed countries, it ranks XX behind cancers of the breast, lung, uterus, and ovaries and XX accounts for 7% of all female cancers. HPV-16 is most often found XX in lesions of the genital mucosa which may have a risk for XX malignant progression. Estimates indicate that HPV-16 has been XX recovered from about 50-60% of all invasive cervical cancers. XX Studies show that the predominance of HPV-16 in high-grade XX anogenital lesions and invasive cancers is evident also for the XX vulva, the penis, and the anus. HPV-16 has also been found in XX approximately 80% of the Bowenoid papulosis lesions (premalignant). XX XX In contrast to the extrachromosomal form of the viral genome found XX in skin carcinomas of EV patients, the viral genome of HPV16 is XX frequently found integrated into the cellular genome in genital XX tract cancers. The integration of the genome requires XX linearization of the genome which most often occurs in the E1/E2 XX region. This results in the inactivation of the E2 repressor of XX the E6 and E7 genes. With repression removed, the E6 and E7 genes XX are able to immortalize keratinocytes. XX XX The DNA of HPV-16 has been molecularly cloned from a genomic XX library of an invasive cervical carcinoma in bateriophage lambda. XX This sequence has been corrected as stated in [3] and [4]; at nt XX 4363 a substitution of a T for an A, and at nt 3906 an additional T, XX respectively. The sense strand is shown, with a numbering system XX matching the first 60 bp of HPV1a, HPV6b and BPV1. The designation XX of the early and late reading frames of HPV-16 is based solely upon XX homology comparison with HPV-6b and BPV-1. Two TATA boxes have been XX identified upstream from the E6 open reading frame, at bp 17 and 65, XX and another upstream from the L1 coding region, at bp 4289. A XX potential CAT box precedes the TATA boxes near the E6 region, at bp XX 7895. Polyadenylation signals can be found at the end of the early XX and late gene regions, at bp 4213 and 7260. Open reading frame XX E4 does not have a translation initiation codon, which is present XX in most other sequenced papilloma viruses. Also, the open reading XX frame for a possible E1 gene product, thought to be involved in DNA XX replication, seems to be interrupted. The author contends that an XX insertion of one nucleotide somewhere in the region between positions XX 1101 and 1168 would generate an E1 open reading frame of normal size.
