XX XX The authors [1] identified the retinoblastoma susceptibility (RB) XX gene encoding a messenger RNA (mRNA) of 4.6 kb on the basis XX of chromosomal location, homozygous deletion, and tumor-specific XX alterations in expression. Transcription of the gene was abnormal XX in 6/6 retinoblastomas while normal in fetal retina and placenta. XX The sequence presented in this entry is derived from human fetal XX retina cDNA. The authors [2] also examined cDNA from the XX retinoblastoma cell line Y79. They found that the mRNA produced XX was a shortened transcript which reflects a deletion of exons 2-6 XX and must have ocurred by a mechanism other than recombination of XX homologous sequences. XX XX The Retinoblastoma (Rb) susceptibility gene maps to chromosome XX 13q14 and encodes a nuclear phosphoprotein of 105kDa. Rb is a XX tumor suppressor gene; its nuclear location and ability to bind DNA XX suggests a role in transcriptional regulation. On the side of the XX protein lies a complementary oncoprotein-binding pocket that is a XX common target for the three viral proteins: E1A of adenovirus, large XX T antigen of SV40, and E7 of HPV. Munger et al. (EMBO 8, 4099-4105) XX determined that the high risk HPV types such as HPV16 and HPV18 bind XX to pRB with higher affinity than low risk types 6b and 11. The amino XX acid locations of the pRB binding site of the HPV16 E7 protein have XX been mapped to a small stretch of amino acids. In addition, pRb has XX shown to associate with a number of cellular host proteins at the XX onco-protein binding site. pRB shows differential phosphorylation XX at different points within the cell cycle.
